Description | Gekweekte umbilikale endoteelselle word meestal gebruik om endoteelselfunksie in vitro te bestudeer. Die nadele hiervan is dat die kweekproses selfunksie nadelig kan beïnvloed en dat die funksie van endoteelselle verskil na gelang van hulle anatomiese ligging. Na aanleiding hiervan het ons ’n vloeikamer ontwikkel om die funksie van vars endoteelselle te bestudeer. Die vloeikamer is so ontwerp dat ’n stukkie bobbejaanaorta (5 cm lank) in 'n geslote vloeibaan geplaas word. Twee stukke bobbejaanaorta is gebruik. Die een is vir 6 ure by 37 °C met Ringer-laktaatoplossing geperfuseer en die ander met dieselfde buffer wat 1 eenheid (E)/ml trombien bevat. Na die perfusie is die perfusate by -70 °C weggevries. Cultured human umbilical cord endothelial cells are mostly used to study the response of endothelium to various stimuli. The effect of the culture process on cell function is however, not known and the function of endothelial cells from different anatomical loci differs. In view of this we developed a flow chamber to study the function of fresh endothelium. The flow chamber was designed to accommodate a five cm piece of baboon aorta in a closed circuit where six hours of perfusion of the aorta with Ringer’s lactate/dextrose buffer at 37 °C was maintained by a peristaltic pump. | |
